However, some reports from Veterinarians and dairy cattle owners, on the negative effects of the vaccine on the health and well-being of the animals and the productivity of their herds, discourage the use of this biological. Based on this problem, this research work was developed to determine the effect of the vaccination process on stress levels and some blood parameters and productive indicators of this in dairy cows. The work was developed in a dairy herd in the Sabana de Bogotá, located at 2,550 m.a.s.l. with daily temperatures between 4 and 20 degrees Cº. For the study, 40 adult cows in production were randomly selected and were assigned in the same way, randomly, to 5 treatments: intramuscular vaccination in the hindquarters, subcutaneous vaccination in the neck table, intramuscular saline injection in the hindquarters, injection of saline solution in the neck table and a control group subjected to pen management, but without any injection. Albumin, globulin, C-reactive protein and Cortisol levels were analyzed at 2 hours and 72 hours after treatment and milk production from 48 hours before treatment to 5 days after. The results showed a higher level of plasma cortisol in the cows vaccinated with subcutaneous injection (p <0.05), a marked increase in C-reactive protein and globulin at 72 hours for the intramuscular and subcutaneous vaccine (p <0.05). The milk production did not have significant changes during the sampling time. It is concluded that vaccination by the subcutaneous route in the neck table generates a greater stress, than intramuscular vaccination in the hindquarters.El mantener el estatus de Colombia como país libre de Aftosa por vacunación implica enormes esfuerzos gubernamentales de la mano de las asociaciones ganaderas y los laboratorios productores de la vacuna. Sin embargo, algunos reportes de Médicos Veterinarios y propietarios de ganado de leche, sobre efectos negativos de la vacuna en la salud y bienestar de los animales y la productividad de sus hatos, desestimulan el uso de este biológico. Basados en esta problemática, se desarrolló este trabajo de investigación para determinar el efecto del proceso de vacunación sobre los niveles de estrés y algunos parámetros sanguíneos y productivos, indicadores de este en vacas lecheras. El trabajo se desarrolló en un hato lechero de la Sabana de Bogotá, ubicado a 2550 m.s.n.m. con temperaturas diarias entre 4 y 20 grados Cº. Para el estudio se seleccionaron 40 vacas adultas en producción en forma aleatoria y se asignaron de igual manera, aleatoriamente, a 5 de tratamientos: vacunación intramuscular en cuarto trasero, vacunación subcutánea en la tabla del cuello, inyección de solución salina intramuscular en cuarto trasero, inyección de solución salina en la tabla del cuello y un grupo control sometido al manejo de corral, pero sin inyección alguna. Se analizaron niveles de albúmina, globulina, proteína C reactiva y Cortisol a las 2 horas y las 72 horas posteriores al tratamiento y la producción de leche diaria, desde 48 horas antes del tratamiento hasta 5 días después. Los resultados mostraron un mayor nivel de cortisol plasmático en las vacas vacunadas con inyección subcutánea (p<0.05), un aumento marcado de proteína C reactiva y de globulina a las 72 horas para la vacuna intramuscular y subcutánea (p< 0.05). La producción de leche no tuvo cambios significativos durante el tiempo de muestreo.
